Targeting oxidative stress at the cellular level

The antioxidant activity of Centextra™ was evaluated by measuring intracellular ROS production in fibroblast cells exposed to oxidative stress. The results showed that the extract significantly reduced intracellular ROS levels compared to control cells, in a dose-dependent way. Moreover, the effect of Centextra™ was stronger than the one observed after exposure to unformulated Centella asiatica extract at comparable concentrations (Fig.1).1

Supporting a healthy inflammatory response

The effect of Centextra™ on the inflammatory response was evaluated by measuring intracellular IL-6 concentration, a key pro-inflammatory marker, in fibroblast cells exposed to a pro-inflammatory substance (lipopolysaccharide, LPS). The data showed that Centextra™ significantly reduced IL-6 levels compared to control fibroblasts. Also in this case, Centextra™ performed better than the unformulated Centella asiatica extract at comparable concentrations.1

Building the foundation for skin matrix repair potential

The in vitro scratch assay is a testing model, used to assess cell migration and wound healing by simulating the closure of a gap in a fibroblast cell monolayer. This method was successfully used to verify Centextra™’s potential for skin matrix repair. In particular, the formulated extract significantly promoted cell migration and monolayer repair, outperforming the unformulated Centella asiatica extract at comparable concentrations: after 24 hours, Centextra™ achieved a wound closure rate of approximately 40% (Fig.2).1

Safeguarding collagen for healthier skin

Collagen is a key structural protein of the dermal extracellular matrix, and plays a crucial role in maintaining the strength, elasticity, and integrity of the skin. With aging and under conditions of oxidative or inflammatory stress, collagen synthesis decreases while at the same time its degradation increases, leading to reduced firmness, loss of elasticity, and impaired skin repair capacity.
The results of the study presented below indicate that Centextra™ helps support collagen homeostasis, restoring collagen levels toward physiological conditions and modulating collagenase activity (Fig.3).1
